The confocal microscope

The Leica SP8 model installed at LIPhy is an instrument shared between the partner laboratories of Labex Tec21, and accessible to external users.
It allows to visualize thick samples with a three-dimensional resolution.

  • Contrast modes: fluorescence (labeled samples), phase contrast or reflection.
  • Objectives 10x to 60x, minimum theoretical resolution 230nm.
  • 7 excitation wavelengths available: 458, 476, 488, 496, 514, 561, 633nm.
  • 4 confocal detectors, 2 of which are ultra-sensitive, 1 transmission detector. Possibility of photon counting.
  • Continuous spectral detection range between 400 and 800nm.
  • Autofocus and 3D motorization for multi-position or mosaic XYZT acquisitions.
  • Possibility to combine complex acquisition sequences ("data live mode").
  • Temperature control, CO2 + humidity insert for acquisitions on cells/tissues.

The Amira software

The microscope is associated with an image processing platform using the Amira software.

  • 3D+T reconstruction and rendering, image or film capture
  • segmentation, tracking, calculation of 3D deformation fields
  • possibility of exporting the analyzed data to Matlab

Free of charge for LIPhy users of contributing teams, 10€/h for other Tec21 member laboratories, 50€/h for external users
